There are MetroCard vending machines near where you would catch the buses.
The cards' vending machines are conveniently located at the bus stop, but if you have heavy luggage or more than two pieces, taking the bus and subway is a very bad idea. While more expensive, use a taxi. Or if you are headed to a midtown hotel, check to see if they have a free courtesy bus from the airport to the hotel.
It is not very easy to get from LGA to Manhattan. You can take a bus, but it takes a long time. You can also take a bus to the subway, but you will have to manage stairs with your bag. If possible, I would take an Uber, Lyft, or taxi.
