If you want to do this at a reasonable cost, take the bus that goes from Newark Airport to Times Square.
First check with the hotel, as some major hotels have a courtesy bus taking their guests to and from the hotel and airport. If they do not do this, you can take a taxi (last time I looked this was $50 plus toll and tip). You can take a bus to the PATH train in Newark, and the train into Manhattan although this will leave you no closer than about 12 blocks from the hotel. There's also a bus from the airport to the Times Square area.
